Question: start with BAM file

Hi, I have RNASeq data mapped outside. indeed I have both FASQ files and BAM files. is it possible to start with BAM files for differential gene expression in galaxy? how?

how to upload large file size? I am unable to upload a single file with file size greater that 2GB?
