I am trying to set up Mothur toolsuite from jjohnson on my local instance and having some troubles....it says mothur 1.33 was not found in conda. Kindly help.

Use the Make.contigs tool form option "Trim with an oligos file?" set to "yes" and select the dataset from the working history.

The file is just plain text and should be formatted as described in the link you reference. Any text editor could be used. The spaces between the data columns should all be a single tab. Make sure there are no extra blank lines at the top, internally, or at the end of the file.
