Can someone help me and explain to me how to clustering sequences into OTUs with Mothur ? I have ITS2 fungal sequences (Illumina MiSeq 2x300 bp). I followed the Galaxy training for the 16S Microbial analysis with Mothur. Its doing well except the Align.seq I got an error when I tried (TERM environment variable not set) I put my fasta file from unique.seq and a reference file (UNITE_public_mothur_full_01.12.2017.fasta). So I skip this step and I went directly to the Pre-clustering step. Everything works but I skip the step Removal of non-bacterial sequences (because I work with fungal sequences) and the step Assessing error rates based on our mock community. I tried to Get.groups and its works and I now i'm trying Dist.seq command and its not working. I took the fasta file from my Get.groups. I cannot skip this step because for the command Cluster i need colum to the dist output from Dist.seqs.

Thank
